|Laminins, a family of extracellular matrix glycoproteins, are the major noncollagenous constituent of basement membranes. They have been implicated in a wide variety of biological processes including cell adhesion, differentiation, migration, signaling, neurite outgrowth and metastasis. Many of the effects of laminin are mediated through interactions with cell surface receptors. These receptors include members of the integrin family, as well as non-integrin laminin-binding proteins. Laminin Receptor is a high-affinity, non-integrin family, laminin receptor 1. This receptor has been variously called 67 kDa laminin receptor, 37 kDa laminin receptor precursor (37LRP) and p40 ribosome-associated protein. The amino acid sequence of Laminin Receptor is highly conserved through evolution, suggesting a key biological function. It has been observed that the level of the laminin receptor is higher in colon carcinoma tissue and a lung cancer cell line than their normal counterparts. In addition, there is a correlation between the upregulation of this polypeptide in cancer cells and their invasive and metastatic phenotypes (1).|

Flow Cytometry - Anti-67kDa Laminin Receptor antibody [EPR8469]; Overlay histogram showing MCF7 cells stained with TA311054 (red line). The cells were fixed with 4% paraformaldehyde (10 min) and then permeabilized with 0.1% PBS-Tween for 20 min. The cells were then incubated in 1x PBS / 10% normal goat serum / 0.3M glycine to block non-specific protein-protein interactions followed by the antibody for 30 min at 22°C. The secondary antibody used was Alexa Fluor 488 goat anti-rabbit IgG (H&L) at 1/2000 dilution for 30 min at 22°C. Isotype control antibody (black line) was rabbit IgG (monoclonal) (0.1ug/1x10^6 cells) used under the same conditions. Unlabelled sample (blue line) was also used as a control. Acquisition of >5,000 events were collected using a 20mW Argon ion laser (488nm) and 525/30 bandpass filter. This antibody gave a positive signal in MCF7 cells fixed with 80% methanol (5 min)/permeabilized with 0.1% PBS-Tween for 20 min used under the same conditions.
