LIN28A, a promoter of pluripotency, is a developmentally regulated RNA binding protein, which selectively blocks the processing of pri-let-7 miRNAs in embryonic cells (1,2). One of the let-7 family members, let-7g targets and represses LIN28 through four 3'-UTR binding sites. It has been reported that LIN28/let-7 pair has the potential to act as a 'toggle switch' that balances the decision to maintain pluripotency vs. differentiation (3). LIN28 was identified as a negative regulator of miRNA biogenesis and suggested that it may play a central role in blocking miRNA-mediated differentiation in stem cells and in certain cancers (1).
