RACK1, a binding protein of heterotrimeric G protein ß? (Gß?) subunits, is a member of the WD40 repeat protein family that binds to activated protein kinase C (PKC) (1). It functions as an anchoring protein to stabilize PKC at the membrane in an active conformation (2). RACK1 has been shown to interact with a variety of proteins and may act as an adaptor/scaffold protein to orchestrate diverse cellular processes, ranging from signal transduction to cell growth (1). Moreover, it has a well established role in integrin and growth factor signaling. RACK1 can interact with ß1 and ß3 integrin receptors, and it regulates the assembly of protrusions necessary for cell adhesion and migration in an Src-dependent manner. RACK1 scaffolds members of the Erk signaling pathway to promote integrin-mediated activation of MAPKs (3). In addition, it is a ribosomal protein that is located at the back of the 40S head region and makes direct contact with ribosomal RNA (4).
