O(6)-methylguanine-DNA methyltransferase (MGMT) is a DNA repair protein that protects tissues against toxic and carcinogenic effects of alkylating agents (1). It is involved in the cellular defense against the biological effects of O6-methylguanine (O6-MeG) in DNA, and repairs alkylated guanine in DNA; a suicide reaction, because the enzyme is irreversibly inactivated (2). MGMT is ubiquitously expressed in normal human tissues (8) but is overexpressed in all types human tumors, including colon cancer, breast cancer, leukemia, lymphomas, and myeloma. Although tumor cells have many mechanisms of resistance to alkylating agents, the expression of MGMT in tumors has a protective effect against cell death induced by alkylating agents, showing a strong correlation between MGMT activity and tumor drug resistance (3).
