FunctionMucins are high molecular weight glycoproteins that provide a mucus protective layer on epithelial surfaces and are involved in cell-cell interactions, signal transduction, and breast cancer metastasis (1). For example, Mucin-1 (MUC1) modulates signaling in ERK, SRC, NF-kappa-B, and Ras/MAPK pathways (2). The expression of MUC1 inhibits the degradation of erbB1, regulators of cell survival and growth, and E-cadherin (3). When MUC1 forms a complex with beta-catenin, it enters the nucleus to activate T-cell factor/leukocyte enhancing factor 1 transcription factors and gene expression (4). MUC1 is expressed on the apical surface of epithelial cells in the airway passages, breast, and uterus (2). Different isoforms of MUC1 have unique tissue specificities. In particular, the 20-kDa isoform 7 is expressed only on tumor cells (2). The alpha subunit has cell adhesive properties, whereas the beta subunit is involved in cell signaling (2).

Flow Cytometry - Anti-MUC1 [EPR1023] antibody; Overlay histogram showing MCF7 cells stained with TA307444 (red line). The cells were fixed with 80% methanol (5 min) and then permeabilized with 0.1% PBS-Tween for 20 min. The cells were then incubated in 1x PBS / 10% normal goat serum / 0.3M glycine to block non-specific protein-protein interactions followed by the antibody for 30 min at 22°C. The secondary antibody used was Alexa Fluor 488 goat anti-rabbit IgG (H&L) at 1/2000 dilution for 30 min at 22°C. Isotype control antibody (black line) was rabbit IgG (monoclonal) (0.1ug/1x10^6 cells) used under the same conditions. Unlabelled sample (blue line) was also used as a control. Acquisition of >5,000 events were collected using a 20mW Argon ion laser (488nm) and 525/30 bandpass filter. This antibody gave a positive signal in MCF7 cells fixed with 4% paraformaldehyde (10 min)/permeabilized with 0.1% PBS-Tween for 20 min used under the same conditions.
