A phospho specific peptide corresponding to residues surrounding tyrosine 1021 of human PDGF Receptor beta was used as an immunogen. This antibody only detects PDGF Receptor beta phosphorylated at tyrosine 1021.

Human platelet derived growth factor (PDGF) receptor is a receptor tyrosine kinase whose activity induces various signaling cascades leading to cell growth and proliferation. The two major isoforms that exist are PDGF Receptor beta and PDGF Receptor alpha. PDGF Receptor beta can only bind to two PDGF isoforms, PDGF-beta and PDGF-delta (1, 2). Upon ligand binding, the receptor undergoes dimerization, resulting in autophosphorylation to allow activation of cytoplasmic SH2 domain-containing signal transduction molecules such as PI kinase, Grb2, Src, etc. (3). Phosphorylation of the PDGF Receptor at Tyrosine 1021 is required for the stable association of PLC gamma to the receptor's C-terminus, the production of inositol phosphates, and initiation of the maximal mitogenic response (4). A chromosomal aberration involving the PDGF Receptor beta is found in a form of chronic myelomonocytic leukemia (5).
