|S100-A9 (MRP14) is a member of the S100 family proteins containing two EF-hand calcium-binding motifs (1). S100 proteins are localized in the cytoplasm and/or nucleus of a wide range of cells, and involved in the regulation of a number of cellular processes such as cell cycle progression and differentiation (2). S100-A9 forms a heterodimer, Calprotectin, with S100-A8 in a calcium-dependent manner. Calprotectin is a major calcuim- and zinc-binding protein in the cytosol of neutrophils, monocytes, and keratinocytes (3). S100-A9 may function in the inhibition of casein kinase and altered expression of this protein is associated with the disease cystic fibrosis (2). |

Immunocytochemistry/ Immunofluorescence - Anti-S100A9 antibody [EPR3555]; ICC/IF image of TA307398 stained DU145 cells. The cells were 4% paraformaldehyde fixed (10 min) and then incubated in 1%BSA / 10% normal goat serum / 0.3M glycine in 0.1% PBS-Tween for 1h to permeabilise the cells and block non-specific protein-protein interactions. The cells were then incubated with the antibody overnight at +4Â°C. The secondary antibody (green) was , DyLight? 488 goat anti-rabbit IgM (H+L) used at a 1/250 dilution for 1h. Alexa Fluor? 594 WGA was used to label plasma membranes (red) at a 1/200 dilution for 1h. DAPI was used to stain the cell nuclei (blue) at a concentration of 1.43ÂµM.
