|CIP4 (Cdc42-interacting protein 4) is a Cdc42 effector that is required for the translocation of GLUT4 to the plasma membrane in response to insulin signaling. It is involved in coordinating membrane tabulation, with reorganization of the actin cytoskeleton during endocytosis. CIP4 binds to lipids such as phosphatidylinositol 4,5-bisphosphate and phosphatidylserine, promoting membrane invagination and the formation of tubules. It promotes Cdc42-induced actin polymerization by recruiting WASL/N-WASP, which in turn activates the Arp2/3 complex. Actin polymerization may promote the fission of membrane tubules to form endocytic vesicles. CIP4 is also required for the formation of podosomes, actin-rich adhesion structures specific to monocyte-derived cells. Furthermore, CIP4 interacts with GTP-bound RHOQ. It is expressed in the brain, colon, heart, kidney, and other tissues (1).|
