Functionp53 acts as both a tumor-suppressor and transcription factor that, upon activation by DNA damage and other cellular stress signals, leads to the transcription of genes triggering cell-cycle arrest, apoptosis, and DNA repair. Various serine/threonine kinases (ATM, p38, Chk) has been limplicated to p53 phospholyation. Phospholyation at serine 392 has been linked to p53 activation. Phospholyation on serine 392 also induces p53 interaction with NF kappa beta - p65 in mouse and inhibition with Cdc2 and MDM2 in human (1,2). The gene for the nuclear phosphoprotein p53 is the most commonly mutated gene yet identified in human cancers. p53 is frequently mutated or inactivated in about 60% of cancers.
