|The heat shock response is transcriptionally regulated by an evolutionarily conserved protein termed heat shock factor (HSF). It appears that there is a human heat shock factor gene family and that at least two separate but related HSF proteins regulate the stress response in humans (1). HSF2, one of the heat shock transcription factors in mammalian cells, is localized to the cytoplasm during normal growth and moves to the nucleus upon activation. Heat shock transcription factors in metazoans contain four hydrophobic heptad repeat sequences, three in the amino terminus and one in the carboxy terminus, which are predicted to form alpha-helical coiled-coil structures analogous to the leucine zipper. It is believed that zipper domains can regulate subcellular localization (2). The promoter regions of some genes, such as the stress-inducible hsp70i gene that codes for a heat shock protein, remain uncompacted, a phenomenon called bookmarking.
