Gab1 (Grb2-associated binder-1) is an adaptor protein that serves to activate numerous intracellular signaling pathways via its interaction with receptor tyrosine kinases (RTKs). Gab1 is a substrate of various RTKs, including EGFR, insulin receptor, cytokine receptors, and B cell antigen receptors; and it acts as a docking protein for several SH2-containing proteins (1-3). Tyrosine-phosphorylated Gab1 may recruit cytosolic signaling proteins to the cellular membrane, where they can act on membrane-bound targets as well as activate downstream pathways (4)
