|Mediates the voltage-dependent sodium ion permeability of excitable membranes. Assuming opened or closed conformations in response to the voltage difference across the membrane, the protein forms a sodium-selective channel through which Na(+) ions may pass in accordance with their electrochemical gradient. It is a tetrodotoxin-sensitive Na(+) channel isoform. Plays a role in pain mechanisms, especially in the development of inflammatory pain (from SwissProt).|

Western Blot: Nav1.7 Antibody - Western Blot was performed using affinity purified SEQer SCN9a antibody. The lanes contain 5-30ug of a whole cell extract. Final concentration of antibodies = 0.1ug/ml (1:10,000 dilution). The blot was probed overnight with the SEQer SCN9a antibody. Blot was then washed according to protocol and probed with goat-anti-Rabbit-HRP conjugate at 1:5000 dilution, washed and developed using chemiluminescence (film exposure 5-30sec). The protein was detected as represented by the band shown.
Immunohistochemistry: Nav1.7 Antibody - Moderate to strong cytoplasmic positivity was seen in most normal tissues. Glandular cells of the gastro-intestinal tract were mostly strongly stained. Strong positivity was also seen in exocrine pancreas, decidual and trophoblastic cells of placenta and neuronal cells in hippocampus and lateral ventricle. Squamous epithelia were weakly positive or negative. Weak to moderate cytoplasmic staining was observed in most malignant tissues. Isolated cases of colo-rectal, endometrial, prostate and cervical cancers were strongly positive. Skin and breast cancers were mainly negative.
