|The elongation of primed DNA templates by DNA polymerase delta and epsilon requires the action of the accessory proteins, proliferating cell nuclear antigen (PCNA) and replication factor C (RFC). RFC, also called activator 1, is a protein complex consisting of five distinct subunits of 145, 40, 38, 37, and 36.5 kD. This gene encodes the 40 kD subunit, which has been shown to be responsible for binding ATP. Deletion of this gene has been associated with Williams syndrome. HEK293T cells were transfected with the pCMV6-ENTRY control (Left lane) or pCMV6-ENTRY RFC2 (RC201138, Right lane) cDNA for 48 hrs and lysed. Equivalent amounts of cell lysates (5 ug per lane) were separated by SDS-PAGE and immunoblotted with anti-RFC2.
Western blot analysis of extracts (35ug) from 9 different cell lines by usin g anti-RFC2 monoclonal antibody (HepG2: human; HeLa: human; SVT2: mouse; A549: human; COS7: monkey; Jurkat: human; MDCK: canine; PC12: rat; MCF7: human).
HEK293T cells transfected with either RC201138 overexpress plasmid(Red) or empty vector control plasmid(Blue) were immunostained by anti-RFC2 antibody(TA504956), and then analyzed by flow cytometry.
