RanBP3 is a protein with a RanBD1 domain that is found in both the nucleus and cytoplasm. It plays a role in nuclear export as part of a heteromeric complex (1). RanBP3 binds to the Crm1 transport receptor and inhibits the binding of unloaded Crm1 to the NPC. In addition, it increases the affinity of Crm1 for both Ran-GTP and nuclear export sequences. In this manner, RanBP3 coordinates efficient cargo loading and RanGTP binding (2). Moreover, RanBP3 has been shown to bind to RCC1 in a Ran-dependent manner and to increase the nucleotide exchange activity of RCC1, resulting in high concentrations of RanGTP in the vicinity of RCC1. Consequently, RanBP3 acts as a scaffold protein by which components of the export complex are concentrated around the RCC1 site, thus promoting complex assembly (3).
