|C-Jun N-terminal kinases (JNK) are distantly related to mitogen-activated protein kinases (ERKs) and are activated by dual phosphorylation on Tyr and Thr. The JNK protein kinase group consists of three isoforms (JNK1, 2, and 3) and regulates transcription activity of the AP-1 complex, and release and regulation of cytochome c (1). JNK becomes activated in vivo in response to pro-inflammatory cytokines or cellular stresses. Its full activation requires the phosphorylation of a threonine and a tyrosine residue in a Thr-Pro-Tyr motif, which can be catalysed by the protein kinases mitogen-activated protein kinase kinase MKK4 and MKK7 (2). Unlike other JNK isoforms, JNK3 is only expressed in the heart and nervous system. JNK3 has been implicated in stress-induced neuronal apoptosis (3).|

Flow Cytometry - Anti-JNK1 + JNK2 + JNK3(phospho T183+T183+T221) antibody [EPR5693]; Overlay histogram showing HeLa cells stained with TA310637 (red line). The cells were fixed with 80% methanol (5 min) and then permeabilized with 0.1% PBS-Tween for 20 min. The cells were then incubated in 1x PBS / 10% normal goat serum / 0.3M glycine to block non-specific protein-protein interactions followed by the antibody for 30 min at 22Â°C. The secondary antibody used was Alexa Fluor? 488 goat anti-rabbit IgG (H+L) at 1/2000 dilution for 30 min at 22Â°C. Isotype control antibody (black line) was rabbit IgG (monoclonal) (1Âµg/1x10^6 cells) used under the same conditions. Unlabelled sample (blue line) was also used as a control. Acquisition of >5,000 events were collected using a 20mW Argon ion laser (488nm) and 525/30 bandpass filter.
