FunctionThe 60-kDa Heat Shock Protein (Hsp60) is a highly conserved member of the molecular chaperone network commonly imported into the mitochondria to assist in the folding and assembly of mitochondrial proteins (1). Under environmental stress conditions, synthesis of Hsp60 is up-regulated then involved in gene expression regulation, DNA replication, signal transduction, differentiation, apoptosis, cellular senescence or immortalization (2). Hsp60 is also believed to protect mitochondrial ATPase activity under conditions of increasing temperature (3). Hsp60, along with Hsp70, has also been shown to play a role in a danger signalling cascade in response to bacterial infection (4).

Immunofluorescent analysis of 4% paraformaldehyde-fixed HeLa (Human cervix adenocarcinoma) cells labeling Hsp60 with TA300692 at 1/150 dilution. The cells were permeabilised with 0.1% Triton X-100. Goat anti-rabbit IgG (Alexa Fluor® 488) (ab150077) at 1/500 dilution was used as the secondary antibody (green). The confocal image shows cytoplasmic staining on HeLa cell line. The nuclear counter stain is DAPI (blue). Tubulin is detected with ab7291 (anti-Tubulin mouse mAb) at 1/1000 and ab150120 (AlexaFluor®594 Goat anti-Mouse secondary) at 1/500 dilution (red). The negative controls are as follows; 1.
