GRB7 (Growth factor receptor-bound protein 7) belongs to a small family of adaptor proteins that are known to interact with a number of receptor tyrosine kinases and signaling molecules. It interacts with epidermal growth factor receptor (EGFR) and ephrin receptors. The protein also plays a role in the integrin signaling pathway and cell migration by binding with focal adhesion kinase (FAK) (1). Best known for its regulatory role in cell migration and tumor metastasis, GRB7 regulates inflammation by coupling NF-kappaB-inducing kinase with erbB/EGFR family receptors (2). The role of Grb7 in these processes depends upon binding to membrane-associated tyrosine kinases through its C-terminal SH2 domain (2). Aberrant overexpression of GRB7 has been found in numerous human cancers, including ovarian cancer (3).
