|D(1A) dopamine receptor (DRD1) belongs to the G-protein coupled receptor 1 family and it is one of the five types of dopamine receptors (D1-D5) (1). DRD1 is the most expressed dopamine receptor in central nervous system and it is involved in the regulation of neuronal growth & development, and modulation of dopamine receptor D2-mediated events. G protein coupled DRD1 activates adenylyl cyclase which leads to activation of cyclic AMP dependent protein kinases. Polymorphism of DRD1 gene has been implicated in bipolar I disorder and attention deficit hyperactivity disorder (ADHD) (1,2). |

Western blot - Dopamine Receptor D1 antibody [EP1560Y]; Anti-Dopamine Receptor D1 antibody [EP1560Y] at 1/20000 dilution + SH-SY5Y cell lysate at 10 ug.Secondary.HRP labelled goat anti rabbit at 1/2000 dilution.Predicted band size : 49 kDa.Observed band size : 75 kDa .
Flow Cytometry - Anti-Dopamine Receptor D1 antibody [EP1560Y]; Overlay histogram showing SH-SY5Y cells stained with TA301064 (red line). The cells were fixed with 80% methanol (5 min) and then permeabilized with 0.1% PBS-Tween for 20 min. The cells were then incubated in 1x PBS / 10% normal goat serum / 0.3M glycine to block non-specific protein-protein interactions followed by the antibody for 30 min at 22°C. The secondary antibody used was Alexa Fluor 488 goat anti-rabbit IgG (H&L) at 1/2000 dilution for 30 min at 22°C. Isotype control antibody (black line) was rabbit IgG (monoclonal) (0.1ug/1x10^6 cells) used under the same conditions. Unlabelled sample (blue line) was also used as a control. Acquisition of >5,000 events were collected using a 20mW Argon ion laser (488nm) and 525/30 bandpass filter. This antibody gave a positive signal in SH-SY5Y cells fixed with 4% paraformaldehyde (10 min)/permeabilized with 0.1% PBS-Tween for 20 min used under the same conditions.
