FunctionThe creatine kinases are a family of enzymes with a highly conserved protein sequence, which are involved in the maintenance of ATP at sites of cellular work .Three cytoplasmic (non-mitochondrial) isoenzymes of creatine kinase (CK)' are readily identified in human tissue. These isoenzymes are dimeric molecules with two dissociable subunits designated as M- (muscle) or B- (brain) type that can reassociate to form the electrophoretically distinct MM, BB, or MB isotypes (1).The protein encoded by creatine kinase B is a cytoplasmic enzyme involved in energy homeostasis. The encoded protein reversibly catalyzes the transfer of phosphate between ATP and various phosphogens such as creatine phosphate (2). Creatine kinase isoenzymes are crucial to energy metabolism, particularly in tissues with high energy requirements, such as skeletal muscle, heart, brain and spermatozoa (3).

Flow Cytometry-Anti-Creatine kinase B type antibody [EPR3927](TA307502); Overlay histogram showing HeLa cells stained with TA307502 (red line). The cells were fixed with 4% paraformaldehyde (10 min) and then permeabilized with 0.1% PBS-Tween for 20 min. The cells were then incubated in 1x PBS / 10% normal goat serum / 0.3M glycine to block non-specific protein-protein interactions followed by the antibody for 30 min at 22°C. The secondary antibody used was DyLight 488 goat anti-rabbit IgG (H+L) at 1/500 dilution for 30 min at 22°C. Isotype control antibody (black line) was rabbit IgG (monoclonal) (1ug/1x10^6 cells) used under the same conditions. Acquisition of >5,000 events was performed.
