|Chromogranin A (CgA, CHGA) belongs to a family of acidic secretory proteins expressed in majority of neuroendroine tissues. Specifically located in the neuroendocrine secretory vesicles, CgA is co-secreted with various peptide hormones and neurotransmitters in neuroendocrine cells. CgA is involved in biogenesis of dense-core secretory granules and immunity to microbes (1). Cleaved specifically by endogenous PC1 (prohormone convertases 1) and furin in vivo, its function has been linked to intracellular hormones and neuropeptides production (2). The measurement of plasma CgA level has been linked to the diagnosis in carcinoid tumors, neuroblastoma, islet cell tumors, phenochromocytoma, and in certain cases for SCLC and prostate cancer (3, 4).|

Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded sections) - Anti-Chromogranin A antibody; Chromogranin A staining on Monkey Pancreas sections. The sections were subjected to heat mediated epitope retrieval using citric acid (pH 6) and blocked using 1% BSA for 10 mins at 21Â°C. TA300689 was used at a dilution of 1/750 using TBS/BSA/Azide for 2 hours at 21Â°C. (Red arrowheads indicate extra Islet positive cells).
Immunocytochemistry/ Immunofluorescence - Anti-Chromogranin A antibody; ICC/IF image of TA300689 stainedPANC-1 cells. The cells were 100%methanol fixed (5 min) and then incubated in 1%BSA / 10% normalgoat serum / 0.3M glycine in 0.1% PBS-Tween for 1h to permeabilise the cells and block non-specific protein-protein interactions. The cells were then incubated with the antibody overnight at +4Â°C. The secondary antibody (green) was , DyLight? 488goat anti-rabbit IgG (H+L) used at a 1/250 dilution for 1h. Alexa Fluor? 594 WGA was used to label plasma membranes (red) at a 1/200 dilution for 1h. DAPI was used to stain the cell nuclei (blue) at a concentration of 1.43uM.
