FunctionThis gene encodes a member of the cysteine-aspartic acid protease (caspase) family. Sequential activation of caspases plays a central role in the execution-phase of cell apoptosis. Caspases exist as inactive proenzymes which undergo proteolytic processing at conserved aspartic residues to produce two subunits, large and small, that dimerize to form the active enzyme. This protein is processed by caspase APAF1; this step is thought to be one of the earliest in the caspase activation cascade. Alternative splicing results in two transcript variants which encode different isoforms. [provided by RefSeq].

Immunohistochemistry (Frozen sections) - Caspase 9 antibody [E23]; TA303419 staining Caspase 9 in rat brain tissue by Immunohistochemistry (Frozen sections).Tissue was fixed in formaldehyde, blocked using 1.5% serum for 1 hour at 22°C and then incubated with TA303419 at a 1/250 dilution for 16 hours at 4°C. The secondary used was an Alexa Fluor 594 conjugated goat polyclonal used at a 1/250.

The secondary antibody used was DyLight? 488 goat anti-rabbit IgG (H+L) at 1/500 dilution for 30 min at 22°C. Isotype control antibody (black line) was rabbit monoclonal IgG (1µg/1x10^6 cells) used under the same conditions. Acquisition of >5,000 events was performed. This antibody gave a positive signal in K562 cells fixed with 4% paraformaldehyde (10 min)/permeabilized with 0.1% PBS-Tween used under the same conditions.
