Vantage™ microRNA Labeling Kit

Marligen’s Vantage™ microRNA Labeling Kit utilizes a proprietary labeling technology to provide exceptional sensitivity for the detection of endogenous microRNAs. This method can label any RNA sample including total RNA, enriched low molecular weight (LMW) RNA, amplified senseRNA and degraded RNA. The simple protocol consists of just two steps. The 3’end of the RNA is poly(A) tailed followed by the ligation of biotinylated 3DNA™ dendrimers containing 15 biotins to provide signal amplification.

Advantages:

  • Labeling can be completed in 45 minutes
  • Highly sensitive labeling method requires as little as 100ng total RNA
  • Biotin dendrimer containing fifteen biotins per microRNA amplifies the signal
  • LMW RNA enrichment is not necessary
  • Will Label degraded RNA from FFPE samples
  • Biotinylated RNA is suitable for multiple applications
  • Compatible for use with xMAP® microRNA profiling kits

Vantage Workflow

This simple protocol consists of just two steps. The 3’end of the RNA is poly(A) tailed followed by the ligation of biotinylated 3DNA™ dendrimers containing 15 biotins to provide signal amplification.

Highly sensitive labeling method requires little input RNA

Titration of Synthetic miR-210

Marligen’s Vantage™ microRNA labeling and detection kits can detect <1pg (or <250 attomoles) synthetic microRNA. In this example, the limit of detection of miR-210 is 0.09 pg (12 attomoles) of synthetic miR.

Labels degraded RNA

microRNA Levels directly Detected in Degrated Total RNA

Total RNA was extracted from normal and tumor lung tissue. The total RNA from both samples was determined to be degraded when analyzed using Agilent LabChips®. The RNA samples were subsequently labeled and detected using OriGene’s Vantage™ microRNA Labeling Kit and Oncology Detection Panel 1 (Cat. No.AM100045). A subset of microRNAs detected by the Vantage™ Oncology Panel 1 is shown here. The data demonstrates that expression profiles from degraded RNA can be analyzed using the Marligen Vantage™ labeling and detection method.
