|The NMDA (N-methyl D-aspartate) receptor in the brain plays a key role in synaptic plasticity, synaptogenesis, excitotoxicity, memory acquisition and learning (1). It mediates neuronal functions in glutamate neurotransmission and is involved in the cell surface targeting of NMDA receptors (2). NMDAR is composed of at least one obligatory NMDA receptor 1 (NR1) subunit and one or more NR2A-D subunits. NMDA is probably regulated by the C-terminal phosphorylation of an isoform of NR1 by PKC and dephosphorylated on Ser-897 by protein phosphatase 2A (PPP2CB) (2). Its phosphorylated state is influenced by the formation of the NMDAR-PPP2CB complex and NMDAR channel activity (2). A role for NMDAR1 in the molecular pathology underlying Huntington disease (HD) has been proposed on the basis of neurochemical studies in HD and the ability of NMDAR1 to mediate neuronal cell death (3).|
